The Sentosa SX system combines reversible adsorption of nucleic acids with the convenient handling of magnetic beads.
Nucleic acids are adsorbed on the magnetic beads. During magnetic extraction, the beads are retained in the deepwell plate while non-DNA components of the samples are pipetted out of the plate. After extraction, DNA is eluted from the beads, aspirated and transferred to a new plate. Eluted DNA is then ready for downstream process.
The extraction procedure is safe and generates reproducible results. It is a 4-step procedure, (1) lysis of samples, (2) adsorption of DNA on magnetic beads, (3) washing of all non-DNA components, (4) elution of DNA from the beads. Steps 2 to 4 are performed automatically by the Sentosa SX101 instrument.
The Sentosa SX Bacteria gDNA Kit v2.0 is used for magnetic beads-based genomic DNA extraction from bacteria in biological specimens on the Sentosa SX101 instrument.
