The use of Sentosa SX Virus Total Nucleic Acid Plus II Kit on the Sentosa SX system combines reversible adsorption of nucleic acids with the convenient handling of magnetic beads to facilitate nucleic acid extraction.

Nucleic acids are adsorbed on the magnetic beads. During magnetic extraction, the beads are retained in the deep well plate while non-nucleic acid components of the samples are removed from the plate. After extraction, nucleic acid is eluted from the beads, aspirated and transferred to a new plate. Eluted nucleic acid is then ready for downstream process.

The extraction procedure is safe and generates reproducible results. It is a 4-step procedure, (1) lysis of samples, (2) adsorption of nucleic acid on magnetic beads, (3) washing of all non-nucleic acid components, (4) elution of nucleic acid from the beads. The entire extraction procedure is performed automatically by the Sentosa SX101 instrument.
